Don’t sound desperate, show a little personality, be clear on what role you’re after and why you would be an asset.
Chances are they won’t have an opening but if your email is memorable, they’ll remember you when that changes. Just don’t overdo it.
Just doing a half decent one puts you in the top 25% of applicants so don't put that much stress on yourself. At least half of people don't do any and another quarter are outright horrible.
The best ones I get include at least some reason why you're interested in that specific agency/boss and some sign that you've done research at more than a cursory level. You should try to include detail about something unique you bring to the table that's relevant to the agency/boss.
Include a specific ask if possible or appropriate (use good judgment) and be concise with all of the above. A little cute/clever helps, but a little goes a long way. Don't overdo it

I was once told that including your Title and Portfolio in the subject line would help. But idk man

Just copy/paste this and put your URL underneath it. It’s a perfect unexpected honest email and you’ll get more clicks.
I was told don’t kill your self on the cover letter, it’s just skimmed. I googled examples of ad cover letters and customized them
