What’s their turnover rate/employee retention and why do they think it is where it is.

Ask if you are replacing someone, or if this a new role. If you’re replacing someone, ask if they are still in the company (got promoted, changed departments) or if they left.
If this is a new role, ask who took on the responsibilities prior to creating a position.
The best answers are that someone got promoted, or the department is expanding. It’s a round about way of asking about attrition.

Good point. I suppose it would depend on how high the position is/how close to the CEO you’ll be. If VP (possibly Director) yes.
The startup I’m at is less than 50, but I just re-read that yours is 300. Big difference after 100.
It's the CEO. Strategic vision and culture would be at the top of my inquiry list. Long term strategy and growth plans, medium and short term goals, values, "how they do business" or establish expectations about work requirements.
Turnover seems blunt. All of those items are important to know and should help inform your go/no-go decision.
What depts are hiring, what's their short term and long term goals for the company? See if that aligns with your position and what you like to do
Weakness/area for improvements for the team your role would be on and company wide? Why did the CEO take their role with the company?
