How difficult is it to get your visa sponsored for a PM/Strategy & Ops role at a large corp. or a large startup ($10B+)?

Currently based in Spain and eyeing a move to the US.

I had my two or three interviews, got an offer for a role at an university, I accepted and am “ hired” . But they’re still checking references . Does that make sense ? I got a update that one of my references did not work out. I provided two other references that I don’t know if they’ll give great reviews due to them being petty. Is it a high chance that they can take the offer away if I don’t meet the requirements for my references ?

I recently stepped away from my position overseeing HR operations at a midsize company (>300 staff). After numerous executive level changes and the departure of our HR VP I was promoted to a role I didn’t particularly want. After managing through it for a year and a half I decided to step down. I’m beginning to look for new opportunities and curious for any tips on getting interviews with larger size companies… Any insider insight?
