Highly dependent on where you are flying from and to and dates. For most direct: London, Madrid - Avios. Paris, Amsterdam - Flying Blue. Flying Blue tends to have a lot of promos for PE and Biz class so I would start there.

Transfer to Virgin and book Delta One to Amsterdam or Paris.
Delta One is their modern business class product and it’s pretty damn good. Similar to Qatar Q suites. You’ll just want to fly the new A350 ones. Check out a few reviews online to get an idea of what it looks like.

Create accounts with a bunch of partners and check award availability for the dates and flights you want. Once you find availability, then transfer points. Transfers from AMEX to most partners are instantaneous. Note that some airlines charge fuel surcharges on award tickets and some don’t. Air Canada can also be an interesting redemption option given very lax routing and stopover rules: you can fly to Asia via Europe and you’re allowed two stopovers. With those rules, you can get a round the world ticket with stops in three places for 150k miles in business class. I did one with stops in Toronto, Koh Samui, and Munich, and also had two 23-hour stops (which don’t count as stopovers) in Bangkok and one in Taipei. From Munich, I bought a separate, inexpensive ticket to Morocco with a stop in Lisbon.

No, and with that many points you should be able to avoid economy on most flights. Virgin is a partner with Delta and ANA, which another person mentioned. Suggest you check out redemption options for both.
