I transferred as an experienced manager and also switched from audit to advisory. Best move I ever made. So timing is fluid...the door is never shut

You have to have 2+ years experience to move from a small firm to Big 4. I did it the minute I had 2 years experience. Also, if you know someone there and the Big 4 firm is offering a 2x experience hire bonus that helps a lot too. Your friends are WAY more likely to help you if they are getting a big bonus out of it.

Doesn't really matter. Find a good fit and jump, if that's what you're looking for.

Seniors are always in the most demand and you need that experience under your belt at the firm you plan to be longer term before you move to manager - your networks and book of business start when you are a senior. You can make the move later, but it will be a harder adjustment

I transferred from a small firm to big 4 within a year of being promoted to senior and transferred as a senior 1. It was fine.
