Say that then, don’t automatically call people cheap
50% for sure or pay the whole thing and they will do it next time. Don’t be cheap
Pay whole amount by yourself! It’s your family that money you spent will be compensated at some point in life!
Who's idea was it to share the room?
You get 50% of the sleeping space, you pay for 50%. But you can ask them and have them make a suggestion how they want to split the cost. Best case you pay for 33%, worst case 50%.

The Brother should cover it all
Especially since it sounds like you shared a room to save money (which is a fine reason), it’s probably ok to ask an open ended question like “hey how should we split the hotel?”. If they offer to cover 66%, great. I would not push the issue if they propose 50-50.
Better yet, if this is a road trip with other costs there’s probably more similar situations (gas, maybe a rental car or some cabs, etc). Set up a Splitwise and figure it out in aggregate.
Disclaimer: I’m not good at these things myself and was brought up with “I pay this time, with the tacit understanding that it’s on them to remember next time”. But I’ve also been a student, so can relate to the conundrum.
Don’t make it about who pays what for vacation. You are relatively better off, just pay whatever and have a good vacation.
Shared a room with a couple for one night once on a layover. I THINK i paid 33% but they paid for the room and I just venmo’d whatever they requested.
