Could be personal preference but Rome was over crowded and somewhat of a let down. The historical things in Rome need to be experienced once in a lifetime but Tuscany and Venice were better areas to visit

Recommend doing a tour of all the landmarks at night. Looks so much better lit up

As long as you're in the city center, there's not a bad location in Rome. Everything is pretty easily seen by walking. All of the sights are pretty tightly centralized. It's a much more manageable city than say Paris. The Vatican is the farthest out if you're staying in the city center. It's still within walking distance, though, or hop on the subway for a quick ride.
What EY1 says. As long as you are in the old city center you are fine. Rome may be my favorite city in the world. Only thing is, it's really, really hot in summer.
