After background check (conflicts or drug test) cleared.

The latter. After anything that could cause the employer to revoke the offer is completed and cleared. (Granted, offers can be revoked even after that, but at least after everything you have to do/clear gets done)
Rising Star
After background check (assuming no complaints do it check) and any drug testing is complete and you have an official start date. Background checks can take a while depending on the company that does them and how far back they look. Safest to make sure everything is completed before giving notice.
I did it after the offer was signed. Had a 90 day notice period and basically no chance of failing the background check.
