I think you’re asking the wrong question. Agencies want to see work that says who you are and what they’ll get working with you. Doesn’t necessarily mean it needs to be a spec, but it should be short.
What kind of Director are you? What’s your voice? It can be a tough question to answer, but if you don’t know, why would an agency want to work with you specifically with all of the options out there.

No specs. Make something worth watching.
One of the main challenges of advertising is delivering a clear and concise messaging for a brand in 30s while making it entertaining and feeling not like an ad. I would say you would need some things in your reel that can prove you can do this. I would be careful making things ‘look as much like an ad as possible’. The majority of ads are complete garbage and depend on common tropes. Make sure you are avoiding these things.
