If you were the one that is presenting the opportunity and you, on your own volition, wanted to move to a different location, then that’s a different conversation…

I would want to fully understand the why behind the required new location, to either know why that particular location is a requirement or to be able to successfully argue that location doesn't matter in the way that they think it does.
Negotiation is about leverage, and your only leverage is whether or not you take the job. You either don't take the job or you change their minds on this requirement.
Unless you’re desperate I’d just decline this offer
