Honestly, do Tokyo/Japan while you can. They are putting some new rules in place to limit tourism starting next year
Of those 3 Tokyo has the most options, Singapore is good for like 2-3 days. Can’t comment on Taipei/Taiwan as much
Taiwan. Last year I did a week tour and visited a good portion of the island.
Osaka - it’s a good food city and close to Tokyo. Seoul is also a good city break, though I’d lean toward Japan

I’ll add: Kyoto is a city of about a million people. You can easily enjoy it as a city as well
How long is the trip for? Singapore for example is good to see but you can cover it all in 2-3 days. Does it have to be 1 country or do you have time to cover a few?
Seoul, Bangkok, KL, Singapore, Bali, Kyoto
Tokyo or Taipei. Or maybe combine Singapore with something else. Hong Kong’s also good for a couple days
if guided tours is relevant to your trip id put osaka/kyoto over Taipei. Take a bike tour in Kyoto. Take the bullet train.
Singapore is good for 2 days, and i dont think you need a tour.
You could stop with Japan and Singapore, or even just Japan.
I've traveled fairly extensively across the world and pretty much everywhere the big cities aren't nearly as cool or fun as the smaller, less traveled cities.
