Yes, that would not be a good look and it would play into exactly the type I would describe as needing improvement in leadership attitude.
What could be helpful, if done well and with humility, would be to explain how you plan to improve based upon that feedback.

I would have a follow-up and ask for more specific feedback, SBI- style of possible. Ask for examples. Talk through where you feel you’ve been displaying leadership and ask about gaps that may still be there. If there truly are no gaps and this is some sort of preconceived notion, that conversation can go a really long way. If there are gaps that might be a bit of a blind spot, you’ll have a more meaningful place to start.

No that would be the worst thing to do and would put you further on that managers list of not being a leader if you sent him an email explaining why he was wrong.
Do some self reflection and really think about why he might have said this. If I were to follow up with him I’d ask for some mentorship on how to improve your leadership attitude. Showing this manager your progressing and making positive changes is what will change his mind, not trying to argue your side.

Ask for specific feedback. Then ask for pointers on how to work on your “weaknesses”. You’d not only gain an ally to champion your cause but you’ll learn as well.
I suggest you write down what you do to support the fact that you have the leadership skills. And ask what else can you do or not do to be a strong leader. In that case you are showing them what you are strong at and also that you want to grow. Defensive approach might bring your manager in the same mode, it won't be as productive in that case. That’s a subjective opinion of a person that probably doesn’t even know how to be a leader in the first place. That places right up there with the 30-yr they gave a ridiculous title to in my company and let play leader. He wouldn’t know leadership if it booped him in the nose.
In all honesty; I just tried to ask for a promotion and ended up going over my main managers head to go to his boss. And it ended very poorly. Unfortunately; you got your answer. He doesn’t think you are fit for that position. You can disagree but the only way to win him; is to prove him wrong. Take the steps and his suggestion and try to improve so next time around he can’t say you don’t deserve that promotion.
It sucks even though I’m sure you deserve that promotion. But it’s only going to make you look bad if you go over his head to his boss. They will think you don’t respect the chain of command.
