No. An unwanted advance is an unwanted advance, regardless of how attractive the person making it is. That would have me feeling anxious too. Also, what planet is your manager from where asking for your number like that is "just being friendly"? It might be in some circumstances, but this isn't one of them. It seems pretty clear he's interested in more than friendship.
I don't think you are overreacting at all. When you are at the desk, you can't just walk away, so any unwanted attention feels bigger. If he is friendly, great, but he still put you in a spot by asking for your number while you were working. You don't owe him anything beyond basic politeness.
If it were me, I would think that either you were not in a serious relationship, you were nothing more than a big tease, or you were into cheating. Either way, I would either tell him the truth and move forward with him OR tell him to move on.
