I signal before interview, signal again prior to letter. Sometimes they'll tell you what the letter has - no point of generating the offer if it is below expectations so I'll tell them they need to change it. That way they have time gathering approvals and sign offs. Offer comes if it looks good signed
You can do both, but the HR person had to get that offer letter approved by a couple people, so renegotiating means they have to go through that again. Ask upfront for any info on the range of comp for you rank/geography/skill, and try to come with your own data.
