We'll read cover letters, but it tends to work in reverse order. Traditionally, the cover letter was the intro to the resume. Now the resume seems to be considered first, and if the person seems like a possible fit the cover letter will be read. The cover letter, if written well, will highlight things not obvious from the resume, and will show some personality that probably wouldn't come through on a resume.
If a job application doesn't ask for a cover letter, I never submit one. I have had many recruiters tell me that they don't read them. I would do one if they ask for one because they are likely using that to narrow things down.
