I make what I need to make. I make $28.78 and I know others make $30-$31 within the company. What should I do?

I am project manager in clinical research industry (CRO). After earning PhD, I gave up my science dream to take a more stable job. After slowly climbing the ladders, I feel like I finally got a seat at the table. Half the days I like the hustle, but now I miss being creative. Received an offer today from Twilio for a L3 program manager (non tech) for 125 base, 60k equity, and a 15k sign on (essentially 140k TC). I currently make more than this and have offers higher than this on the table. Anyone have insight on how much I can counter?

Graduating Ph.D. program 2021, received offer from tiny boutique for 140TC+random small boutique benefits. Would be better commute, non metro area closer to home. Waiting to hear back from well known life science boutique (think ZS, Clearview, Putnam). Offer would be for NYC office, would likely relocate.

If offer is given, a) at what $ is this worth and b) is brand everything for first firm out of school?
