Be prepared to walk away. Might sound scary but it's a powerful negotiation tactic. If the company isn't willing to meet your expectations, be prepared to move on. This doesn't mean you have to burn bridges but it shows you're confident and serious about your worth.
Of course you can. You should go for it. If they value you, you should be compensated accordingly.
You can always negotiate. They may say no but it never hurts to ask. I would decide what number you want to make and then ask for a little above that so they can negotiate a little back and forth unless they just say no from the jump.
