Yeah that will probs be tough. They are budgeting your headcount in that office and your hours, so I would not do that. Also, the Charlotte office might have a chip on their shoulder about the offices in "hot" cities, so an SF request will probably not go over well.

It's really just practice dependent. A lot of practices are not city focused, or even region focused.

Obviously you can ask.
The range of possible outcomes stretches from getting what you ask for to causing issues to getting your offer pulled.
You can defs ask, if you get a fast no, move on.
The opportunity is probably a very good one, even if you don't like the city. I lived in a city for a while that wasn't anywhere near my top choice, I then grinded out results and then made a transfer.

Depends on if the practice is regional or national. National practice there may be some opportunities to have a different location, unsure what would happen with the offer value.
If it's a regional practice, it may not be possible because the other region may not have staffing needs. Still worth asking though.
You can ask. Asking by itself won't cause problems, or at least they'll tell you before any problems occur. Don't worry about your offer being pulled. Most likely outcome is that they say "can't change the offer now, but you can explore a transfer after joining".
