I typically refrain from asking for anything specific - tends to limit what they offer (and their "make it go away" reaction is usually a paltry 500 starpoints). Try calmly stating what you found and then follow it up with "I've been <placed into an apparently unsanitary room> - what is the hotel going to do to make me feel comfortable about staying here tonight and in the future?" Then you can decide what combination of upgrade, points, free meal, drink coupons, minibar credit, etc., will keep you as a happy customer. Usually works best to avoid saying "you" so the desk person doesn't take it personally. Good luck!
