Keep in mind that typically, if you've already applied outside of a referral, then someone cannot actually refer you in the system. Same thing if someone has already referred you, someone else can't.
Tdlr: referrals help but make sure you find the best person possible to refer you (right practice, right level, etc)

Yes, if the referer knows you and advocates for you. If they are more senior in the org, it will help you get that first interview, even if there isn't an obvious fit.
