Got an offer to go from contractor to firm member (entry level) , the job was customized for me, how much % (industry standard) can/should I negotiate up from the offered salary?

I’m having a super hard time finding a job in pharma R&D, all the jobs available seem to be analytical - I have 4 years experience working in a lab (3yrs at a CRO & 1yr working as an associate scientist on a drug product development team). I got an offer for a senior research associate ($60k) but I currently am an associate scientist in MD making $75k. Do R&D pharma jobs exist in the Denver/Boulder area? Is it me or are the salaries low because everyone wants to live in Denver?

I do things (late nights/weekends) that I don't think my managers/partner knows about since everything is running well - directly correlated with elected hours spent. Is it weird to bring this up?
