Stop submitting resumes. Reach out to people likely to be involved directly via LinkedIn. You’re competing with literally hundreds of people. You’ll have to do more to stand out, beyond a cover letter rewrite IMO.

"Applying" is a good thing to do, and keep doing it.
BUT, that shouldn't be the main way you try to find a job in advertising. And if it's the only way you're going about it, prepare to be disappointed for a long time.
Reach out. Get to know people. Ask for informational interviews. Contact people you know. Ask them if they know anyone you could talk to. Work it.
Youve got to change your strategy. Your CV needs a rewrite, hire someone for $200 and just do it!
Finding 100 open roles you want to apply to is actually encouraging as that’s a lot of opportunities but are you customizing each resume you submit to the job description? Are you looking for common connections at the company to see if they can pass in your resume or at the very least give you a referral link? Modify your approach to quality over quantity! Hang in there
I applied to literally 150+ places and got only three responses—Ogilvy and two tangentially-related jobs. Been working at Ogilvy since.
Could being the type of person to jump to giving up have anything to do with why 100 jobs have passed right by that resume?

Good point. OP, make sure to take “Jumps to giving up after only 100 tries” off your resume. As D1 pointed out, it may be turning recruiters off

I was at one of the large holdcos a year ago. It was already all juniors doing the work.

Applying isn’t a great strategy. Agencies are hunting for people. With specific skills or reputations. Just sending out resumes is a waste of time. You should be networking, creating and making “your brand” shine.
Your resume probably needs a facelift. See if a friend would do a rewrite or if you can glance at her’s. You can also hire a resume writer. Lastly, customize your CL. I know it’s a lot of work but start from scratch each time and experiment with different tones. A trained eye can see a standard CL with a few changed words and will think you’re lazy.
Be daring. Call that company, and always have as many irons on the fire as possible. Don’t ever rely on interviews that are going well even if you’re in the final rounds. Keep at it until you get your first paycheck. It just takes one.
Most reciters are dicks, factor that in.
It’s true. Recruiters see the $ over our heads before our best interests.
Don’t give up but take a step back and reassess how you have approached your applications: same resume for all, same cover letter for all, submissions through job boards ...?? That’s the assessment you should do.
Once you have done this assessment, you should understand what to change. In any case, lean on your network, prepare a short summary of your experience including one/two sentence(s) highlighting what you could bring to an organization but ... don’t give up
