Do everyone irrespective of their experience needs to prepare for DSA? Or they just want the frontend developer to be proficient in HTML, CSS and JS just like they mention in JD. And also do they consider people from Tier 3 colleges?

Which round of interview is it? If it's a first round, then they probably won't bring up salary, or if they do, it'll be to share a range or ask you what your desired range is. I always say it's negotiable, depending on the offer, and wait to see what they offer before negotiating (if necessary).
I had a phone screening now I have the zoom interview. They asked what my desired salary was and I told then I was open to discuss it. They mentioned what the role will pay during the screening but what they offered and my research on the compensation doesn't equivalate. My thoughts on that is because I'm a recent graduate. So I'm not sure how to go about asking.
I ask what the budget for the role is and they usually give a range! Just casually if they ask if you have questions just be like “oh! Yes actually, what is the budget for this role looking like?” and I get a good answer most of the time. If they’re not willing to answer I find it odd.
This is what I do as well. And then I usually ask for a little above the range. Always aim higher than where you actually want to land because there will usually be a little back and forth.

I second M2's comment. By asking for the budget for the role, you'll have an idea if there would still be some room for negotiation. If there is, then you can negotiate within the reasonable range. Good luck!
