How do Healthcare /Pharma corporations do project prioritisation within the CIO’s group given there are limited resources and more than enough transformation project requests ?

I accepted a $115K base salary, 13% bonus, 10k sign on bonus (scientist supervisor) job and two weeks later, got a $135K base, 17% bonus, and 15K sign on bonus (Senior scientist role) offer from another company. Should I accept the new offer ? How do I tell the previous company about this? Also, I’ve already negotiated with the first company and the $115K base is the negotiated salary. TC: $223k for 1st year (156k base, $64k sign on for 1st year and $50k for second year, with 46 shares - vesting 5% on 1st year anniversary, 15% on 2nd year and 20% every 6months on 3rd and 4th year).
