For me personally, I knew when I no longer felt fulfilled. I started dreading going to work every day, and watched the clock until it was time to go home. When it basically became drudgery, I knew it was time to explore other avenues.

Aside from self fulfillment some measurable things to look for include difficultly with or denial when making attempts to discuss anything relating to career succession, merit increases, bonuses and benefits, etc. this should not be an uncomfortable discussion and it should be easy to measure.
