Apply through the online job portal and be referred by someone in the company (if you have the connection). Most importantly tailor your resume for the exact job you want. The first round is HR weeding out the resumes that were too generic. When there's 400 applications, they are going to be quick to sort who gets to move forward and who doesn't.
Also when cover letters are stated as "optional", write the letter.
Director1's advice is good. Tailor application, get a referral if you can, and include a pithy, clear and tailored cover letter.

I did fire off some cold LinkedIn messages tonight but don’t have high hopes
Not sure if this is helpful, but all of the interviews I’ve had at big companies have come from recruiters reaching out to me on Linkedin. Make sure your experience has bullet points, not just companies and titles (use your resume bullets). Also make sure to update your summary. Both of these changes will help you appear in searches
Referrals are your best luck unfortunately. Second best would be stalking LinkedIn to see if hiring managers post about open roles on their team.
