Wait until you’ve already taken on that responsibility for a while (at least 6 months) then you can point to your fantastic performance at your new responsibilities, as well as the added value you provided, as the justification for the raise.
I second CD1. You have to put some time under your belt with those added responsibilities with proof of performance.
I’ll add that you should ask for clear direction on the expectations and responsibilities of the next level. Have a conversation with your new manager and let them know you want to work towards the next level. Ask what you need to work on to get there.

And by love, I mean hate.
Exactly. For all you know, they are hauling ass to find a replacement supervisor for you because they know you’re not ready or capable to take that job on yourself, and they’re worried about what might go wrong while you’re relatively unsupervised. Marching in and demanding a raise when you’ve been covering the job for two days is going to come off incredibly clueless and tone-deaf.
Do it, do it well, use this as rationale for asking for a promotion once you’ve demonstrated some success. You’ll either get a raise then or you can take your new skills to a new job.
If you’re truly underpaid AND you have the ability to do the next job up, you could just get a new job right now. If you don’t feel like that would work (because you don’t have the work history or experience to get a more senior job yet), you probably have your answer.

Are you taking on things for the long term or are they eventually backfilling the role?
As far as I know, it’s both. I’ll be taking on a decent amount and they’re hiring someone to do the other parts of their job as well as some other duties that the person leaving wasn’t doing
