What do you know that your brother doesn’t? He’s a grown adult - my assumption would be he just doesn’t care. And telling him what to do isn’t going to change his mind, just cause tensions.
Personally, I wouldn’t say anything. But if you do, I recommend approaching it from a place a curiosity instead of telling him what to do.

Unless you want to give up your spot on the guest list to make room for whoever you think just has to be invited, then say nothing. Immediate family (assuming they are on speaking terms and not such a screwup they might ruin the day), their close friends. Then extended family and wider friend circle.... not necessaily in that order. His budget, his list.
Rule of thumb, if your mom isn't crying over who is excluded, no intervention is needed. If she is, that means she tried to intervene and he said sorry. In which case your intervention will also fail.

I would honestly just let him do his thing because at the end of the day it is his wedding and his budget. I get what you are saying and you could gently suggest it but after that I would probably drop it.
I wouldn't say anything. If he knows of their existence, then he chose not to invite them. If he doesn't know of their existence, then they're clearly not close.
I did not invite many family members and it was fine. How those people react is their own responsibility 🤷♀️
